If a card, such as Faramir, Prince of Ithilien, gains defender+1 through bearing a possession, and then a Shadow card discards this possession, what happens? Does one of the minions skirmishing Faramir have to be reassigned?

Defender +1 is only in any use when you are assigning. So, from what I understand of your question, if during a skirmish involving Faramir and 2 minions, his possession is discarded, he would still resolve the skirmish with the 2 minions.
