Can I use Southron Troop to reduce a fellowship from 9 companions to six companions?
(By this I mean can I use an assignment action multiple times if the conditions for that assignment action remain the same)

As long as you spot 7 companions you can repeat the assign option

I've once played vs someone who said I could only use assignment actions once... he said I couldn't do that and I believe him.
But... if you can perform the action twice, it means you could technically assign Gorgoroth Assassin to companion X bearing Desperate Defense of the Ring and then re-assign the same Gorgoroth Assassin to a companion Y who bears another Desperate Defense of the Ring and then re-assign him again to companion X and continue until the RB is corrupted.

Many assignment actions assign a minion to a companion. You cannot do this unless both of them are unassigned.

So as long as both are unassigned you can do this, but since assignment special abilities go first, you get to do this as long as you can meet the requirements:

So Southron Troop is ok because discarding a companion cancels the assignment action on Frodo and then both Southron Troop and Frodo are unassigned, allowing the assignment action to trigger again (should there still be at least 7 companions).

Exactly.
Think of Saruman, Servant of the Eye. As long as you can still exert (the requirements are met), you can use his assignment action again. As long as you have 7 companions and Southron Troop is not yet assigned, you can repeat.
